Encapsulation of Allergens into Core–Shell Chitosan Microparticles for Allergen-Specific Subcutaneous Immunotherapy

IgE-mediated allergic reaction occurs in response to harmless environmental compounds, such as tree and grass pollen, fragments of household microorganisms, etc. To date, the only way treat allergy is allergen-specific immunotherapy (ASIT), which consists a prolonged subcutaneous administration allergen extracts or recombinant proteins. The long duration treatment, cost risk life-threatening adverse reactions are main limiting factors for ASIT. aim this work was develop proteins encapsulated chitosan-based microparticles that can be safely administered at high doses rash protocol. egg white allergen, Gal d 1 protein, used model antigen. protein packed into core–shell type (MPs), core formed with succinyl chitosan conjugated 1, subsequently coated shell by quaternized chitosan. obtained MPs containing (Gal-MPs) were non-toxic macrophage fibroblast cell lines. At same time, Gal-MPs quickly engulfed bone marrow-derived dendritic cells RAW264.7 cells, visualized using flow cytometry confocal microscopy. Encapsulated not recognized 1-specific IgE ELISA. Female BALB/c mice immunized subcutaneously three times week 2 weeks. Immunization resulted IgG titers 1250 ± 200 without production. Allergy control vaccinated induced low-dose injections withers mice. control-sensitized but Thus, preventive vaccination allergens safe rapid; it significantly reduces production respiratory oral allergens.
